What is Ipamorelin?

Ipamorelin is a research peptide that stimulates the release of growth hormone (GH). It is composed of five amino acids and is thus known as a pentapeptide. Ipamorelin is often lumped together with CJC 1295, though they are two different compounds.

Ipamorelin was originally developed by Novo Nordisk Inc. It was investigated in phase 2 trials as a treatment for postoperative ileus, a condition in which the bowel stops functioning after surgical procedures. However, due to lack of efficacy, it was discontinued. [R]

Pharmacological profiling using growth hormone-releasing peptide (GHRP) and growth-hormone-releasing hormone (GHRH) antagonists has clearly demonstrated that Ipamorelin stimulates GH release via a GH-like receptor. Its selectivity for GH is similar to that displayed by GHRH.

 

How does Ipamorelin Work?

Ipamorelin mimics the actions of ghrelin, a gut hormone that affects food intake, fat deposition, and GH release. It binds to the ghrelin receptor (or the GH secretagogue receptor, GHSR) in the brain to selectively stimulate the release of GH from the pituitary gland. [R] [R]

Research has shown that GH secretion goes down with age. The decrease in lean body mass and increase in adipose tissue that occurs with aging has been partly attributed to the age-associated decline in GH secretion and insulin-like growth factor-1 (IGF-1). [R]

 

Ipamorelin Research

Multiple animal studies have been conducted on Ipamorelin. Phase 2 clinical trials were conducted to study the ghrelin mimetic Ipamorelin for the management of postoperative ileus in bowel resection patients. Let’s look at the research carried out on Ipamorelin to date.

 

Ipamorelin and Weight Gain

A 9-week animal study was conducted to assess ipamorelin’s effects on adiposity and weight gain. The study involved female GH-deficient and GH-intact mice. After 9 weeks, body weight in the GH-deficient mice increased by 15.3% in those treated with Ipamorelin. [R]

It is worth noting, however, that there were no changes in the relative weight of the dissected organs with Ipamorelin, while GH led to increased liver weight. This suggests that patients treated with ipamorelin may not experience the same organomegaly as seen in GH-treated patients.

 

Ipamorelin and Bone Health

Ipamorelin and GH-releasing peptide-6 increased bone mineral content in adult female rats. As measured in vivo, both Ipamorelin and GHRP-6 increased total, tibial, and vertebral bone mineral content compared with vehicle. All substances also increased the tibial area bone mineral density. [R]

In another study, Ipamorelin dose-dependently increased longitudinal bone growth rate (LGR) in adult female rats. There was also a pronounced and dose-dependent effect on body weight gain. [R]

 

Ipamorelin and Glucocorticoids

Glucocorticoids (GCs) are naturally-occurring steroids. They perform many functions in the body, including interrupting inflammation and treating conditions caused by inflammation. However, their side effects include weight gain, water retention, and muscle weakness, among others. [R]

In vivo in an adult rate model, the ability of Ipamorelin to counteract the catabolic effects of GC on skeletal muscles and bones was investigated. A group of 8-month-old female rats was injected under the skin for 3 months with GC (methylprednisolone), GHS (Ipamorelin), or both.

In conclusion, the decrease in muscle strength and bone formation in GC-injected rats was counteracted by simultaneous administration of the selective growth hormone secretagogue. The study supports the therapeutic use of GHSs in patients receiving glucocorticoids. [R]

 

Ipamorelin and Diabetes

Research suggests that Ipamorelin evokes significant increases in insulin secretion from the pancreas of normal and diabetic rats. Ipamorelin stimulates insulin release through the calcium channel and the adrenergic receptor pathways. [R]

The study was conducted to examine the effects of Ipamorelin on insulin secretion from pancreatic tissue fragments of normal and diabetic rats.

 

Ipamorelin and Muscle Growth

A study was conducted to examine the effects of Ipamorelin to counteract the catabolic effects of glucocorticoids on skeletal muscle and bones. It was shown that Ipamorelin counteracted the decrease in muscle strength and bone formation found in GC-injected rats. [R]

Longer-term clinical trials on randomized populations are needed to investigate the potential anabolic effects of this growth hormone secretagogue.
