4-Andro’s main ingredient, 4-Dehydroepiandrosterone (4-DHEA), is a positional isomer of the DHEA compound. The chemical structural modification of DHEA reduces its thermogenic properties (calorie burning) while increasing its androgenic and anabolic properties and endothelial nitric oxide synthase activity by increasing its potential binding affinity to muscular receptors.
A recent study on the physiological and hormonal influence of androstenedione found that during 12 weeks of androstenedione or androstenediol use, a significant increase in the aromatization by-products estrone and estradiol was observed in research subjects. In the dione group, total testosterone levels significantly increased by 16% after 1 month of use, but by the end of 12 weeks, they returned to pre-treatment levels.

What is 4-Andro?
4-Andro, also known as 4 Androstenediol, is a chemical known as prohormone. The body has the ability to convert prohormones into anabolic hormones, which, in turn, help stimulate muscle growth and generate protein synthesis. Some of these anabolic hormones include testosterone. [R]
4-Andro is produced naturally in the body by the adrenal glands, as well as the testes and the ovaries. It acts as a precursor in testosterone and estrogen hormone production. That is to say that the body could potentially use endogenous 4 Andro to produce testosterone and estrogen. [R]

Additionally, it has been found that the conversion rate of 4-Andro to testosterone has massive potential, with research showing a ratio as high as 15.76%. Androstenediol Vs Androstenedione
Androstenediol (4-Andro) and Androstenedione are both precursors of testosterone.
Both are produced by the adrenal glands, as well as the testes and ovaries. In men, the body uses both these hormones to produce testosterone. In women, these hormones act as intermediaries to estrogen, a hormone that gives women feminine characteristics.

Multiple human studies have shown that one of the main ingredients of 4-Andro may possess the potential to increase serum IGF-1 levels in the body. These studies have also found DHEA to have possible positive impacts on knee muscle strength and lumbar back strength in males.
4-Andro and IGF-1
Dehydroepiandrosterone (DHEA) is the main active ingredient of 4-Andro. A study found that administration of a 50mg daily dose of DHEA for 3 months to age-advanced men and women resulted in an elevation (10%) of serum levels of insulin-like growth factor-1 (IGF-1). [R]
The findings from this study led researchers to assess the effect of a larger dose (100 mg) of DHEA for a longer duration (6 months) on circulating sex steroids, body composition, and muscle strength. An increase in serum IGF-1 levels was observed in both genders. [R]
Insulin-like growth factor 1 (IGF-1) is a hormone that manages the effects of growth hormone (GH) in your body. It also mediates the protein anabolic and linear growth-promoting effect of pituitary GH. Together, IGF-1 and GH promote the natural growth of bones and tissues. [R]
4-Andro and Testosterone
Testosterone is an important androgenic growth hormone. It regulates tissue repair, secondary sex characteristics, and various growth functions. In men, it’s thought to regulate sex drive, bone mass, fat distribution, muscle mass and strength, and the production of sperm. [R]
In a study conducted in 2000, the effects of 200 mg/day oral administration of Androstenedione or 4-Andro were observed. It was found that both free and total serum testosterone concentrations were significantly elevated by the 4-week measurement period (p < .05). [R]
Similarly, a 2013 study found that taking 50 mg of DHEA per day raised the free testosterone levels of middle-aged adults undergoing high-intensity interval training (HIIT). Therefore, DHEA supplementation may have significant potential benefits related to HIIT adaptation. [R]
4-Andro and The Potential Implications for Muscle Strength
In the abovementioned study, the effects of 100mg of DHEA on muscle strength were also observed. It was found that in men, but not in women, knee muscle strength 15.063.3% (P ¼0.02), as well as lumbar back strength 13.965.4% (p ¼0.01), increased.
The increase in knee strength in men may be of particular value. That is because knee weakness is strongly correlated to falls and morbidity in the elderly. The mechanism by which the increase occurred in men in response to DHEA is currently unknown.
Furthermore, these differences in response to DHEA administration between males and females may reflect a gender-specific response to DHEA and/or the presence of confounding factor(s) in women, such as estrogen replacement therapy.
4-Andro and Potential Effects on Depression
A recent study set out to investigate whether DHEA is more effective than a placebo in the treatment of depressive symptoms. All the subjects had depression not resulting from other psychiatric conditions or medical comorbidities.
The meta-analysis applied in this review pointed to a significant effect in favor of treatment with DHEA compared to placebo. It concluded that DHEA might be a more effective alternative to the compounds used in the treatment of depression. [R]
In another study, six middle-aged and elderly patients with major depression were openly administered DHEA. Preliminary data from the study suggest DHEA may have antidepressant and pro-memory effects. The data should encourage double-blind trials in depressed patients. [R]
4-Andro and Potential on Weight
DHEA administration has been shown to reduce abdominal visceral fat accumulation and protect against insulin resistance in lab animals. A double-blind, placebo-controlled study was conducted to determine whether DHEA therapy decreases abdominal fat in elderly humans.
Compared with the placebo, the 6 months of DHEA replacement resulted in decreased body weight (2.4kg vs. 0.6kg), with no difference in response between men and women. Significant decreases in abdominal visceral fat occurred during the 6 months of DHEA replacement. [R]
It is not known if DHEA helps younger patients to lose weight.
4-Andro and The Potential Relevance to Mood
A study was carried out to investigate the effects of DHEA administration in healthy young men. DHEA administration led to a reduction in evening cortisol concentrations and improved mood and memory. Recollection accuracy in the episodic memory test was also significantly improved. [R]
Another study demonstrated that DHEA reduced activity in the amygdala and hippocampus, enhanced connectivity between the amygdala and hippocampus, and enhanced activity in the rACC. These results demonstrate that DHEA may potentially reduce activity in regions associated with the generation of negative emotions. [R]
Considering that activity in these regions is altered in mood and anxiety disorders, our results provide initial neuroimaging evidence that DHEA may potentially be useful as a pharmacological intervention for these conditions.
4-Andro and Potential Effect on Skin
Topical DHEA may potentially improve skin brightness and counteracts the papery appearance of skin and epidermal atrophy, a characteristic feature of hormone-related skin aging. Topical DHEA could also potentially act on skin processes related to wrinkles, but this result remains to be confirmed. [R]
Data from another study suggest the possibility that topical DHEA could be used as an efficient and physiological anti-aging skin agent. This study was a placebo-controlled, randomized, prospective study performed with 75 postmenopausal women aged 60-65 years. [R]
A clinical trial was also conducted to determine the response of skin parameters related to skin aging to the local action of DHEA in postmenopausal women. Unfortunately, the trial results are yet to be published, even though the study has been complete since 2008. [R]
4-Andro and The Potential to Aid Sexual Dysfunction
An analysis was conducted to review the contemporary knowledge regarding DHEA and erectile dysfunction (ED). It was observed that low levels of DHEA were associated with a higher risk for ED in men, and low sexual responsiveness in women. [R][R]
However, there were no significant differences in sexual performance outcomes between placebo or DHEA groups in 79 men with sexual dysfunction and androgen deficiency in a more recent study. In a separate study, DHEA improved feelings of well-being but not libido in older males. [R][R]
