MK-677 or Ibutamoren is an anabolic research compound that is popular amongst researchers, especially in animal studies, for its promising potential to effectively increase lean body mass with an aim to grow muscle. Due to the potential ability to increase growth hormone levels, the research aims have shifted towards the potential efficacy of MK-677 supplementation as a means of elevating muscle mass, and muscle strength, and reducing overall body fat.

Studies indicate that MK-677 compounds may spur the growth of new muscle cells by signaling the pituitary gland to create more growth hormones and by stimulating IGF 1 release in the liver. According to studies, these hormones together may increase protein synthesis and strength while simultaneously boosting recovery rate and enhancing nitrogen retention.

In a double-blind randomized controlled trial, 24 obese mice received MK-677 therapy for two months. Results showed their basal metabolic rates and muscle mass increased. Another study on mice found that MK-677’s effects reversed muscle wasting and muscle loss. Meanwhile, another study carried out on mice suffering hip fractures found evidence of improved muscle strength when administering an MK-677 research cycle.

How Does MK 677 Work?

Animal studies have shown that MK 677 mimics the action of ghrelin, the growth hormone (GH) secretagogue that stimulates the release of GH. MK 677 works by attaching to one of the brain’s ghrelin receptors (GHSR) and activating them, increasing GH levels.

Multiple studies have confirmed MK 677’s potential ability to raise GH levels. One study on dogs showed that Ibutamoren elevates GH after oral doses as low as 0.125 mg/kg. Another animal study found that prolonged administration of MK-677 increased peak GH concentrations. [R] [R]

Three double-blind, placebo-controlled clinical studies reached the same conclusion. Subjects were given bodyweight-dependent doses of MK 677 for 2 – 9 weeks. The final results showed a sustained increase in GH levels compared to samples collected at the study’s onset. [R]

MK-677 and SARMs

Despite being a growth hormone secretagogue, MK 677 is often grouped with selective androgen receptor modulators or SARMs. This is mainly because it has produced a somewhat similar set of effects in animal, pre-clinical, and clinical studies.

However, despite producing similar effects, MK 677 is not a SARM. Selective androgen receptor modulators work by binding to androgen receptors, mainly in the muscle and bone tissue. Ibutamoren, as stated above, has the potential to mimic the action of ghrelin in the brain.

If effects are to be considered, Ibutamoren finds its place in the growth hormone-releasing family of peptides. Like Hexarelin, GHRP-2, and GHRP-6, MK 677 attaches to the ghrelin/growth hormone secretagogue receptor to stimulate GH release.

It might be beneficial that Ibutamoren is not a SARM. SARMs may impact natural androgen production and lower testosterone levels. Researchers thus have to take many precautions when administering SARMs to research subjects. MK 677 doesn’t need similar precautions.

MK-677 and Growth Hormone

A study was conducted to determine the effect of MK-677 on growth hormone release in obese subjects. The study involved oral treatment of MK-677 and recruited twenty-four healthy obese subjects. The impact of oral administration of MK 677 on GH secretion was observed.

The final results showed that there was a significant increase in GH levels. Another encouraging sign was that the increase persisted at 2 and 8 weeks of treatment. It was also observed that the initial dose of MK-677 produced a greater increase in GH levels than any of the upcoming doses. [R]

In a 24-month-long placebo-controlled trial, Ibutamoren administration led to increased GH and IGF-1 levels with no severe side effects in research subjects. Researchers demonstrated that this resulted in a potential increase in body weight, a decrease in LDL (“bad”) cholesterol levels, and an increase in bone density. [R]

Similar results were obtained following a two-month treatment of obese subjects with MK-677. Fat-free mass saw a significant increase in the Ibutamoren treatment group, compared to the placebo group. However, no effect on body fat was observed. [R]

Research involving elderly adults found that MK-677 has the potential to stimulate bone turnover and increase bone deposition and formation. This could positively impact bone density, as highlighted by another study in which MK-677 treatment improved gait speed and reduced the number of falls in elderly patients with hip fractures.
