What Is Semax?

Semax is a synthetic analog of the adrenocorticotropic hormone 4-10 fragment (ACTH). This synthetic neuropeptide was initially developed in Russia in the 1980s to potentially prevent and treat circulatory disorders and acute cerebral hypoxia (caused by traumatic brain injury). [R][R]

While Semax is yet to be approved by the FDA, multiple studies have highlighted its potential. For example, Semax has been shown potential in improving memory, boosting mental clarity, and increasing dopamine levels in the brain. It might also possess neuroprotective properties. [R]

How does Semax Work?

Given the fact that it hasn’t been extensively studied outside Russia, the potential mechanisms of action of Semax aren’t well-understood. However, some early research has identified a few possible routes of action, including:

 

1) Increases Brain’s BDNF Levels

Brain-derived neurotrophic factor (BDNF) is a protein produced in the central nervous system. It potentially helps neurons survive by aiding them in their growth, mutation, and development. Aside from that, BDNF may plays a crucial role in synaptic plasticity, responsible for learning and memory. [R]

Evidence from multiple animal studies suggests that Semax can potentially stimulate BDNF synthesis. In one study, nasal application of Semax had potential to increase BDNF levels, both in rat hippocampus and basal forebrain. 2) Reduces Breakdown of Enkephalins

Enkephalins are naturally occurring peptides with potent pain-killing effects. They are primarily produced in the brain and have been implicated in blood pressure regulation, memory processes, neuroprotection, wound repair, respiratory control, and various other functions throughout the body. [R]

A cell-based study has shown that Semax peptide may inhibit enkephalin-degrading enzymes in the human serum. The inhibitory effects of this neuropeptide and Selank (another synthetic peptide) were found to be potentially more potent than other inhibitors of encephalin-degrading enzymes. [R]

 

Semax Research

Multiple clinical trials have been conducted on Semax. This synthetic peptide has also attracted various animal and cell-based studies. 1) Semax and Dopamine

Evidence from an animal study suggests that Semax may activate dopaminergic brain systems in rodents. This effect was mediated by D-Amphetamine. Furthermore, the potential increase in dopamine levels was above 20-fold compared with the basal values. [R]

A 2004 study by the same researchers corroborated the findings from the abovementioned study. This study has shown that intraperitoneal administration of 0.15 or 0.6mg/kg of Semax showed potential to raise the intracellular level of dopamine in the striatum region of rat brains. [R]

 

2) Semax and ADHD

A 2007 paper proposed that Semax may have good therapeutic potential in attention-deficit hyperactivity disorder (ADHD). It drew on several lines of evidence to make this proposal. [R]

First, the paper highlighted how Semax potentially stimulated the processes required for memory and learning formation in adults. It then highlighted various studies showing the potential effects of Semax on BDNF release and how this neuropeptide has modulated brain development in animal studies.

Finally, multiple animal studies were quoted showing how Semax, when used alongside one of the most commonly used agents in the treatment of ADHD, potentially increased the agent’s effects on dopamine levels. Thus highlighting how Semax may improve the therapeutic effects of psychostimulants in AHD.

 

3) Semax and Memory

A 1997 Russian study demonstrated that Semax could have the ability to significantly improve memory and attention in healthy men under extreme conditions. The study also noted that Semax is capable of potentially stimulating operative memory and attention in experimental animals and human beings. [R]

Furthermore, it was shown that the potential improvements in memory and attention remained over a prolonged period (20-24h after intranasal administration in 0.015-0.050mg/kg doses). More research is needed to fully understand and verify the potential effects of this peptide on memory.

In another rat study, Semax prevented the learning and memory inhibition caused by heavy metals. The inhibition-countering effect of Semax was the same or at least a considerable degree as ascorbic acid, confirming that this peptide may hold antioxidant pharmacological properties. [R]

 

4) Semax and Brain Health

A study conducted on rat brain focal ischemia showed that Semax’s neuroprotective effects stem from its ability to affect the expression of genes related to the immune and vascular systems.

In a clinical trial involving 100 patients recovering from an ischemic stroke, the addition of Semax to intensive forms of conventional therapy accelerated the restoration of damaged brain functions, especially motor disorders. The treatment lasted only 5 and 10 days. [R]

In another clinical trial, Semax potentially reduced overall brain damage in people with reduced blood brain flow. This synthetic peptide also probably reduced the risk of future strokes. Furthermore, results from the trial show that Semax was well-tolerated by the study’s participants. [R]

In a small-scale study, the effects of Semax on optic nerve disease were assessed. Results show that adding Semax with other treatments potentially improved the recovery rate. The combination also potentially reduced the disease’s intensity and improved the patient’s visual functions. [R]

 

5) Semax and Heart Health

An animal study was carried out to investigate how the peptide Semax affects the heart rate of rats after myocardial infarction (MI). Results showed that Semax prevented hypertrophy (excessive growth) of the heart caused by hypertension (high blood pressure). [R]

Another rat study established that Semax moderates the hyperactivity of the sympathetic nervous system resulting from MI. If left unchecked, the hyperactivity of the sympathetic nervous system might promote heart failure during the post-infarction (post heart attack) period. [R]

 

6) Semax and Stress

Semax has been shown to potentially counteract some of the effects of stress in animals. This synthetic peptide has demonstrated its potential ability to alleviate stress-induced proliferation of macrophages, types of white blood cells that can become hyper-responsive following exposure to toxicants. 7) Semax and Depression

A single application of Semax led to a maximum 1.4-fold increase in BDNF levels in the rat hippocampus. It also stimulated a 1.6-fold increase of tyrosine kinase B receptor (TrkB. These findings are intriguing in finding a potential therapeutic treatment for depression. [R]
