A recent study on the effects of T3 on rats found that the increased expression of thyroid-hormone receptor beta (T3’s receptor) that is increased during caloric restriction is not abolished by T3 supplementation and that T3 supplementation leads to increased leptin gene expression. What is Liothyronine?
Liothyronine is a synthetic thyroid hormone that is used to treat hypothyroidism and may have some application in enlarged thyroid gland conditions. It is being well-researched as a potential replacement for the body’s own thyroids, which are important for metabolism, and energy levels. T3 (Liothyronine) is the most potent metabolite as compared to Levothyroxine(LT4). Its bioavailability and pharmacokinetics are similar to those of T4 but faster in the onset of action, with a maximum effect of about 24h and passes off in 1 week. [R]
How Does T3 Liothryonine Sodium Work?
The endogenous thyroid hormones are replaced by liothyronine and eventually act physiologically by regulating DNA transcription and protein synthesis. The thyroid hormone liothyronine has been demonstrated to bind to thyroid receptor proteins on DNA specifically. As a result, it has the potential to increase energy expenditure, accelerate the rate of cellular oxidation, stimulate growth, mature, and metabolism of body tissues, aid in nerve myelination and synaptic process development in the nervous system, and improve carbohydrate and protein metabolism.[R]

Numerous animal and human studies have been conducted on the potential application of Liothyronine. Below follows some of the relevant findings:
T3 Liothyronine Sodium on Hypothyroidism
The endocrine system illness known as hypothyroidism, also known as underactive thyroid, low thyroid, or hypothyreosis, is characterized by insufficient thyroid hormone production by the thyroid gland. Many Thyroid Hormones (TH) that occur naturally and synthetic ones have been identified and tested for potential TH (ant-)agonistic or direct pharmacological action over the last century following the discovery of T4, but few only made it to clinical practice. In a study conducted on women, Liothyronine (LT3) therapy improves the quality of life of women with residual hypothyroidism on LT4 or LT4/LT3 combination therapy.
After 12 weeks of LT3 therapy, subjects showed significant improvements in cognitive complaints and experience less tiredness. Short-term LT3 does not induce biochemical or clinical hyperthyroidism, and no cardiovascular adverse effects are recorded. Further studies are needed to evaluate the long-term safety and efficacy of LT3 monotherapy [R]

Combination Therapy of Liothyronine and Levothyroxine
As people continue to search for new medication options for hypothyroidism a study aimed to examine the current knowledge regarding combination therapy with LT4 and LT3. Due to insufficient evidence to recommend combination therapy, it was concluded that levothyroxine therapy alone is the standard of care. [R]
However in a study wherein patients undergoing combination use of LT4 and/or synthetic liothyronine (LT3) therapy showed an improving quality of life and did not have additional side effects of atrial fibrillation, cardiovascular disease, or mortality in patients of all ages with hypothyroidism. Until clear advantages of levothyroxine plus liothyronine are demonstrated, the administration of levothyroxine alone should remain the only potential medication of choice for replacement therapy for hypothyroidism.
T3 Liothyronine Sodium on Fat Loss
As mentioned earlier one effect of Liothyronine is weight loss. A study of 19 subjects with a diet high in unsaturated fatty acids was treated with liothyronine. The results showed that weight loss was induced by the administration of liothyronine and this was evident from an increased metabolism [R].
Skepticism towards the use of LT3 in subjects with hypothyroidism is based on concerns regarding the increased risk of developing potential adverse effects on the heart and skeletal structures, as well as clinical problems caused by variable serum hormone levels. [R]
However, a present study shows that LT3 substitution for LT4 at equivalent doses decreases body weight without detectable differences in cardiovascular function or insulin sensitivity.[R]
Furthermore results from a new study presented at ENDO 2021 may show that older adults with subclinical hypothyroidism may lose more weight and fat mass with liothyronine than levothyroxine.[R]
T3 Liothyronine Sodium on Cold Intolerance
Brown adipose tissue(BAT) function is completely lost in severe hypothyroidism. A key regulator of BAT thermogenesis is thyroid hormone as it is known for its effects on body temperature. Recent study results suggest that LT3 may reduce dermal heat loss. Cold-stimulated skin temperatures significantly decreased in patients receiving LT3 than LT4, suggesting that LT3 may be useful for individuals with hypothyroidism and cold intolerance. Thermography data suggested increased brown adipose tissue(BAT) activation in hypothyroid patients with cold intolerance, but this finding was not corroborated by the assessment of the microRNA biomarker of BAT activation. T3 Liothyronine Sodium on Depression
Liothyronine (T3) augmentation has been a potential treatment of choice for patients with unipolar depression. However, there are many unanswered questions about efficacy and safety, especially for long-term use. Given the proven benefits of T3 in treating depression and its safety profile, it is important to review the existing literature on the use of this medication during the treatment phases. [R]
T3 Liothyronine Sodium on Multiple Sclerosis
Several studies have found that thyroid hormone aids in the promotion of remyelination to treat Multiple Sclerosis(MS).
Thyroid hormone was found to promote remyelination in various animal models via various mechanisms, including neuroprotection of myelin and axons, increased myelin-forming protein and nerve growth factor, and promotion of oligodendrocyte lineage development and differentiation.
In a first phase I clinical trial to investigate the safety, tolerability, and optimal dosing of Liothyronine(LT3) for remyelination in people with MS, L-T3 was shown to be safe and well-tolerated up to an MTD( Maximum Tolerated Dose) of 75 mcg TDD(Total Daily Dose). However, more research is needed for longer periods of Liothyronine use, as the study also has a small sample size, limiting generalisability.[R]
T3 Liothyronine Sodium on Women Subject’s Infertility
T3 Liothyronine Sodium therapy may be helpful because infertility is frequently the only or first sign of either incipient hypothyroidism or metabolic insufficiency.
In a study wherein T3 Liothyronine Sodium therapy was done with 38 female research subjects who had struggled with infertility for a long time, 14 women had given birth after 6 months of therapy, while 10 additional women’s menstrual cycles and ovulation patterns had normalized. Even in women who don’t have visible thyroid issues, liothyronine seems to be a potent medication for increasing female fertility.
The encouraging findings still imply that more research should be done to improve the efficacy of liothyronine in treating infertility.[R]
However, it is noted that researchers found Liothyronine sodium to interfere with birth control pills. Research showed that other hormones simultaneously with thyroid medication can interfere with fertility hormones and as such the drug interactions may potentially prevent absorption. [R]
T3 Liothyronine Sodium on Male Subject’s Fertility (Spermatogenesis)
Treatment with L-thyroxine reduced sperm motility and, in some cases, entirely destroyed it; in contrast, liothyronine increased prospective fertility by increasing the quantity of active sperm without reducing the speed of their movement. However, it is clear that much more research is needed before we can offer Liothyronine to subfertile or relatively fertile males.[R]

Even though shipping took longer than expected, this liquid T3 is totally the right dosage and works wonderfully (most liquid T3s are apparently expired and don’t contain the T3 they should. this one does).
One note is that the dropper could use more graduations for more accurate dosing, instead of every 0.25mL only.
