How Does Sunifiram Work?
Although its exact working mechanism remains unknown, Sunifiram has been shown to:
- Stimulate the release of acetylcholine. In one animal study, Sunifiram reportedly increased acetylcholine release. Another preclinical study showed it blocked the effects of scopolamine, a drug that blocks acetylcholine release. [R][R]
- Activate AMPA receptors. Results from a mice study suggest that Sunifiram activates the AMPA-mediated neurotransmission system. Plus, it may block AMPA receptor activation by drugs that suppress the activity of AMPA receptors. [R]
- Regulate glucose uptake. Authors of a cell-based study report that Sunifiram may increase glucose uptake at a lower dose and increase it at a mega dose. [R]
- Activate NDMA glutamate receptors. Two cell-based studies suggest how Sunifiram may activate NDMA glutamate receptors. That is, by promoting the release of the neurotransmitter glutamate, which is proven to bind to glutamate receptors. [R][R]

Sunifiram and Memory
A preclinical study has reported that Sunifiram partially reversed surgically-induced impairments in memory in mice. It also reduced long-term potentiation. LTP is one of the main mechanisms in synaptic plasticity and plays a vital role in the ability to acquire, encode, and store memories. [R]

In one animal study involving mice, small doses of Sunifiram (0.01 mg/kg of body weight) increased the rate at which mice learned how to avoid shocks. The enhanced learning ability was measured using a behavioral task referred to as the passive avoidance test. [R]
According to another animal study, Sunifiram dosing (0.1 mg/kg body weight) reportedly increased the rate at which mice learned how to recognize social partners after brief interactions.

Sunifiram and Pain Sensitivity
Only one animal study has investigated the effects of Sunifiram on pain sensitivity.
The study investigated how mice responded to heat-related pain after being administered Sunifiram. A medium dose of Sunifiram (0.01 mg/kg) was reported to induce a mild pain-blocking effect in the mice. However, at a high as well as at a low dose, much weaker effects were reported. [R]
Sunifiram and Wakefulness
Evidence from one mouse study indicated that Sunifiram may promote wakefulness. [R]
